Arfaptin 2, also known as ARFIP2, is a Rac1 binding protein necessary for Rac-mediated actin polymerization and the subsequent formation of membrane ruffles and lamellipodia. ARFIP2 has also been shown to interact with the ADP ribosylation factor ARF6, a GTPase that associates with the plasma membrane and intracellular endosome vesicles, in a GTP dependent manner. Arfaptin 2 also regulates the aggregation of mutant Huntingtin protein by possibly impairing proteasome function. Expression of ARFIP2 was shown to be increased at sites of neurodegeneration.
