Chemokines are a group of small (~8-14kD), mostly basic, structurally related molecules that regulate cell trafficking of various types of leukocytes through interactions with a subset of 7-transmembrane, G protein-coupled receptors. Chemokines also play fundamental roles in the development, homeostasis, and function of the immune system, and they have effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis. Chemokines are divided into 2 major subfamilies, CXC and CC. This gene is a CXC member of the chemokine superfamily. Its encoded protein induces a chemotactic response in activated T-cells and is the dominant ligand for CXC receptor-3. The gene encoding this protein contains 4 exons and at least three polyadenylation signals which might reflect cell-specific regulation of expression. IFN-gamma is a potent inducer of transcription of this gene. Sample Type: Serum, plasma, cell culture supernates, tissue homogenates Intended Use: For the quantitative determination of human interferon inducible T-cell chemoattractant (I-TAC) concentrations in serum, plasma, tissue homogenates. Sensitivity: 7.8pg/ml Range: 31.2- 2000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of human I-TAC. No significant cross-reactivity or interference between human I-TAC and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for I-TAC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any I-TAC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for I-TAC is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of I-TAC bound in the initial step. The color development is stopped and the intensity of the color is measured.
