This gene encodes a cell surface adhesion molecule that belongs to a family of adhesion/homing receptors. The encoded protein contains a C-type lectin-like domain, a calcium-binding epidermal growth factor-like domain, and two short complement-like repeats. The gene product is required for binding and subsequent rolling of leucocytes on endothelial cells, facilitating their migration into secondary lymphoid organs and inflammation sites. Single-nucleotide polymorphisms in this gene have been associated with various diseases including immunoglobulin A nephropathy. Alternatively spliced transcript variants have been found for this gene. Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of rat L-selectin concentrations in serum, plasma and tissue homogenates. Sensitivity: 64pg/ml Range: 78.125-5000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of rat L-selectin. No significant cross-reactivity or interference between rat L-selectin and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for L-selectin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any L-selectin present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for L-selectin is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of L-selectin bound in the initial step. The color development is stopped and the intensity of the color is measured.
