This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. This protein is structurally related to the CXC subfamily of cytokines. Members of this subfamily are characterized by two cysteines separated by a single amino acid. This cytokine displays chemotactic activity for monocytes and basophils but not for neutrophils or eosinophils. It has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis and atherosclerosis. It binds to chemokine receptors CCR2 and CCR4. Sample Type: Serum, plasma and tissue homogenates Intended Use: For the quantitative determination of canine monocyte chemotactic protein 1/Monocyte chemotactic and activating factor (MCP-1/MCAF) concentrations in serum, plasma and tissue homogenates. Sensitivity: 0.147ng/ml Range: 0.125-8ng/ml Specificity: This assay has high sensitivity and excellent specificity for detection of canine MCP-1. No significant cross-reactivity or interference between dog MCP-1 and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MCP-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MCP-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MCP-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MCP-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
