Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of human platelet-derived growth factor-BB (PDGF-BB) concentrations in serum, plasma and tissue homogenates. Sensitivity: 3.12pg/ml Range: 31.2-2000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of human PDGF-BB. No significant cross-reactivity or interference between human PDGF-BB and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PDGF-BB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PDGF-BB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PDGF-BB is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PDGF-BB bound in the initial step. The color development is stopped and the intensity of the color is measured.
