This gene encodes a secreted inhibitor which protects epithelial tissues from serine proteases. It is found in various secretions including seminal plasma, cervical mucus, and bronchial secretions, and has affinity for trypsin, leukocyte elastase, and cathepsin G. Its inhibitory effect contributes to the immune response by protecting epithelial surfaces from attack by endogenous proteolytic enzymes, the protein is also thought to have broad-spectrum anti-biotic activity. Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of human secretory leukocyte protease inhibitor (SLPI) concentrations in serum, plasma and tissue homogenates. Sensitivity: 7.81pg/ml Range: 31.25-2000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of human SLPI. No significant cross-reactivity or interference between human SLPI and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SLPI has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SLPI present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SLPI is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SLPI bound in the initial step. The color development is stopped and the intensity of the color is measured.
