This protein is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. This cytokine can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis. Some transcripts that skip the last exon of this gene and continue with the second exon of the neighboring TNFSF13 gene have been identified, such read-through transcripts are contained in GeneID 407977, TNFSF12-TNFSF13. Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of human soluble tumor necrosis factor-like weak inducer of apoptosis (sTWEAK) concentrations in serum, plasma, tissue homogenates. Sensitivity: 7.8pg/ml Range: 31.25-2000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of human sTWEAK. No significant cross-reactivity or interference between human sTWEAK and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for sTWEAK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sTWEAK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for sTWEAK is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of sTWEAK bound in the initial step. The color development is stopped and the intensity of the color is measured.
