This gene is one of several Cys-Cys (CC) cytokine genes clustered on the q arm of chromosome 16. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene displays chemotactic activity for T lymphocytes, but not monocytes or granulocytes. The product of this gene binds to chemokine receptors CCR4 and CCR8. This chemokine plays important roles in T cell development in thymus as well as in trafficking and activation of mature T cells. Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of human thymus activation regulated chemokine (TARC) concentrations in serum, plasma, homogenates. Sensitivity: 3.9pg/ml Range: 15.6-1000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of human TARC. No significant cross-reactivity or interference between human TARC and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TARC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TARC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TARC is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TARC bound in the initial step. The color development is stopped and the intensity of the color is measured.
