Phosphoserine aminotransferase (EC 2.6.1.52, UniProt Q9Y617, also known as Phosphohydroxythreonine aminotransferase, PSAT) is encoded by the PSAT1 (also known as NLS2, PSA, PSATD) gene (Gene ID 29968) in human. PSAT1 is a class-V pyridoxal-phosphate-dependent aminotransferase family member that catalyzes the reversible conversion of 3-phosphohydroxypyruvate and to 1-phosphoserine, as well as the conversion of 3-hydroxy-2-oxo-4-phosphonooxybutanoate to 4-phosphonooxy-L-threonine, using, in both cases, L-glutamate as an amine donor, which is converted to 2-oxoglutarate. PSAT1 is involved in serine biosynthesis from glycolytic intermediates and PSAT1 upregulation is reported in non-small cell lung cancer (NSCLC) and colon cancer cells, while PSAT1 knockdoown is shown to suppress the proliferation of MDA-MB-468 and BT-20 breast cancer cells. Applications: Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilution: Immunocytochemistry: 1:250, detects PSAT1 in A431, HeLa, HepG2, and NIH/3T3 cells. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
