HVCN1 is a voltage-gated proton channel highly expressed in immune tissues. Channels like HVCN1 mediate the proton conductances required by phagocytic leukocytes for the oxidative burst that underlies microbial killing. High expression in spleen, whole blood, bone marrow, and macrophages, with weak expression in all other tissues examined. The deduced 273aa protein contains 4 transmembrane segments that correspond to the voltage-sensing domain of other ion channels, but it lacks transmembrane segments characteristic of a pore domain. Database analysis showed enrichment of HV1 in immune tissues, such as lymph node, B lymphocytes, monocytes, and spleen. RNA dot blot analysis detected highest expression in lymph node. Western blot analysis revealed a 32kD HV1 protein in most immune tissue examined. Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:100-1:5000 Western Blot: 1:50-400 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 Immunocytochemistry: 1:50-500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
