Transforming growth factor, beta-induced, 68kD, also known as TGFBI (initially called BIGH3, BIG- H3), is a protein which in humans is encoded by the TGFBI gene. It is mapped to 5q31.1. This gene encodes an RGD-containing protein that binds to type I, II and IV collagens. The RGD motif is found in many extracellular matrix proteins modulating cell adhesion and serves as a ligand recognition sequence for several integrins. This protein plays a role in cell-collagen interactions and may be involved in endochondrial bone formation in cartilage. It is induced by transforming growth factor- beta and acts to inhibit cell adhesion. Sample Type: Cell lysates, cell culture supernates, serum, plasma (Heparin or EDTA). Intended Use: Sandwich High Sensitivity BioAssay(TM) ELISA kit for Quantitative Detection of Rat Beta IG-H3/TGFBI. Sensitivity: <15pg/ml Range: 156pg/ml-10,000pg/ml Specificity: Recognizes natural and recombinant Rat Beta IG-H3/TGFBI. There is no detectable cross-reactivity with other relevant proteins. Test Principle: This Rat Beta IG-H3/TGFBI BioAssay(TM) ELISA Kit was based on standard sandwich enzyme-linked immune- sorbent assay technology. A specific for beta IG-H3/TGFBI has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: G24-G685) and test samples are added to the wells, a biotinylated detection specific for Beta IG-H3/TGFBI is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Rat Beta IG-H3/TGFBI amount of sample captured in plate.
