This assay kit uses a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of OxLDL in human serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Sensitivity: 0.52ng/ml Range: 1.23-100ng/ml Specificity: This assay has high sensitivity and excellent specificity for detection of Oxidized Low Density Lipoprotein (OxLDL). No significant cross-reactivity or interference between Oxidized Low Density Lipoprotein (OxLDL) and analogues was observed. Precision: Intra-Assay: CV<10% Inter-Assay: CV<12% Test Principle: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to OxLDL has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled OxLDL and unlabeled OxLDL (Standards or samples) with the pre-coated antibody specific to OxLDL. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of OxLDL in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of OxLDL in the sample. Kit Components: 96-well strip plate, 1x Plate Standard, 2x vial Detection Reagent A, 1*120ul Detection Reagent B, 1*120ul TMB Substrate, 1*9ml Wash Buffer (30X), 1*20ml Standard Diluent, 1*20ml Assay Diluent A, 1*12ml Assay Diluent B, 1*12ml Stop Solution, 1x6ml Storage and Stability: Store microtiter strips, Standard, Detection Reagent A, Detection Reagent B and positive control at -20C. After reconstitution of the postive control store at 4C and use within 5 days. Store all the other components at 4C. Unused kit is stable for 6 months after receipt. Once kit components are opened, it is highly recommended to use remaining reagents within 1 month provided this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.
