IDO catalyzes the first and rate-limiting step in the main pathway of human tryptophan catabolism, the kynurenine pathway. Proinflammatory mediators, such as endotoxin and IFN-gamma induce the expression of IDO in several tissues. IDO-dependent suppression of T cell responses might function as natural immunoregulatory mechanism. Physiological IDO activity has been implicated in T cell tolerance to tumors, dysfunctional selftolerance in non-obese diabetic (NOD) mice, and as a protective negative regulator in autoimmune disorders. Source: Recombinant protein corresponding to aa2-403 from human IDO1, fused to His-Avi-Tag at N-terminal, expressed in E. coli. Molecular Weight: ~48kD Applications: Suitable for the use in study of enzyme kinetics, substrate specificity, and screening inhibitors. Other applications not tested. Assay Conditions: The assay was performed by UV absorption using a recombinant IDO1 or TDO2 and L-Tryptophan substrate. The UV absorption signal at 321nm is correlated with the amount of N-formylkynurenine reaction product of reaction. All of the reactions were conducted at room temperature. The 100ul reaction mixture in IDO or TDO Assay Buffer contains ~40nM IDO1 (200ng/well for 71182 or 210ng/well for 79081) or varying concentrations (0-320nM) of IDO1, 900 tryptophan, and the coupled reaction components. The reaction mixture incubated for 165min respectively while reading the UV absorption signal. For the negative control (blank), assay buffer was added instead of the enzyme. Absorption signals were measured using a Tecan Infinite M1000 plate reader. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
