The mature recombinant human BMP-10, generated by the proteolytic removal of the signal peptide and the propeptide, is a disulfide-linked homodimeric protein. Based on N-terminal sequencing, the mature recombinant human BMP-10 starts at Asn 317. The monomer has a calculated molecular mass of 12.2kD and migrates as an approximately 12.2kD protein in SDS-PAGE under reducing conditions. Source: Recombinant corresponding to Asn317-Arg424 from human BMP10 expressed in CHO cells (O95393). Activity: Measured by its ability to induce alkaline phosphatase synthesis by MC3T3E1 osteoblastic cells (Mazerbourg, S. et al., 2005, J. Biol. Chem. 280:32122). The ED50 for this effect is typically 15-60ng/mL. Reconstitution: It is recommended that sterile 4mM HCI containing at least 0.1% human serum albumin or bovine serum albumin be added to the vial to prepare a stock solution of no less than 10ug/mL. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile 4mM HCI, with 0.1% HSA or BSA. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
