In cardiac tissue brain natriuretic peptide (BNP) is synthesized as 134aa precursor (prepro-BNP), which is cleaved by proteases to form a 26aa signal peptide and a 108aa pro-BNP. Proteolytic digestion of pro-BNP results in formation of 76aa amino-terminal NT-proBNP and biologically active 32aa BNP hormone molecule. Both proBNP and NTproBNP circulate in human plasma and have been proposed as markers for early diagnosis of left ventricular dysfunction as well as prognostic markers of possible cardiac complications at patients with heart failure. Applications: Suitable for use in ELISA. Other applications have not been tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Specificity: Immunoreactivity is confirmed by reaction with monoclonal antibodies specific to different fragments of NT-proBNP molecule. Predicted molecular mass 8.6kD confirmed by MALDI-MS. Contains an additional methionine residue at the N-terminus (in comparison with native NT-proBNP). Does not contain a fusion partner. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
