| Calyculin A, a polyene phosphate, was initially isolated from the marine sponge Discodermia calyx. It is a very potent, cell-permeable PP-1 and PP-2 inhibitor (IC50: 2nM and 0.5-1nM respectively, and extremely specific for PP-1 and PP2A classes of protein serine/threonine phosphatase among other acidic, alkaline and protein phosphatases. For 10uM stock, reconstitute in 1ml DMSO. Store in aliquots at -20C in the dark. Treat cells with 50-100nM calyculin A for 5-30 minutes. Calyculin A must first be dissolved in an organic solvent such as DMSO, ethanol, acetone or methanol. The recommended solvent for stock solutions to be used for biological experiments is DMSO.
