Synthetic phosphopeptide corresponding to residues surrounding Ser33, Ser37 and Thr41 of human beta-catenin. Species sequence homology: porcine, chicken, bovine, Xenopus, zebrafish and canine
Beta-catenin is a key downstream effector in the Wnt signaling pathway. It is implicated in two major biological processes of vertebrates: early embryonic de- velopment and tumorigenesis. CK1 phosphorylates b-catenin on Ser45. This phosphorylation event primes b-catenin for subsequent phosphorylation by GSK-3. GSK-3b destabilizes b-catenin by phosphorylating it at Ser33, 37 and Thr41. Mutations in these phosphorylation sites, which result in the stabilization of b-catenin protein levels, have been found in many tumor cell lines. Applications: Suitable for use in Western Blot.. Other applications have not been tested. Recommended Dilutions: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
