Immunogen: Synthetic peptide corresponding to residues with the heavy chain subunit of human Cathepsin D.
Cathepsin D is a ubituitiously expressed lysosomal aspartyl protease involved in the normal degradation of proteins (1). It is synthesized as an inactive 43kD preprocathepsin D that is cleaved and glycosylated to form a 46kD procathepsin D and then further cleaved to produce 28kD and 15kD subunits (heavy and light chains, respectively) (2). Cathepsin D may also be secreted into the cytosol during apoptosis and contribute to cleavage of substrates implicated in the apoptotic pathway (3). Numerous studies have suggested that cathepsin D plays a role in neuronal degradation and malignant transformation, particularily in breast cancer (4-9). Applications: Suitable for use in Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Immunohistochemistry (Paraffin): 1:200 Flow Cytometry: 1:200 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
