Procathepsin L isolated from the human lung cancer cell line EPLC 32M1.
Cathepsin L is a lysosomal cysteine proteinase that plays a major role in intracellular protein catabolism. Its substrates include collagen and elastin, as well as alpha 1 protease inhibitor, a major controlling element of neutrophil elastase activity. Cathepsin L has been implicated in several pathologic processes, including rheumatoid arthritis, muscular dystrophy, Alzheimers, cancer, myofibril necrosis in myopathies and in myocardial ischemia, and in the renal tubular response to proteinuria. Cathepsin L, which is a member of the peptidase C1 family, is a dimer composed of disulfide linked heavy and light chains, both produced from a single protein precursor. At least two transcript variants encoding the same protein have been found for its gene. Application: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 2ug/ml Immunohistochemistry (Frozen/acetone fixed): 4ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
