CD45 or Leukocyte Common Antigen (LCA/T200) family is comprised of at least four isoforms (CD45RA, CD45RB, CD45RC, and CD45RO) of membrane glycoproteins with MW of 180kD-220kD. It consists of a large, heavily glycosylated, amino-terminal extracellular domain of 400-550 amino acids, a single transmembrane region and a large carboxy-terminal cytoplasmic domain of 700 amino acids. CD45/LCA is expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues. The intracellular portion of CD45 proteins has phosphatase activity and is involved in regulation of transmembrane signals, functioning as a leukocyte membrane receptor capable of transducing signals via its phosphotyrosine phosphatase domains. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: Neat, 10ul labels 10e6 cells in 100ul Optimal dilutions to be determined by the researcher. Hybridoma: Spleen cells from immunised DA rats were fused with cells of the Y3/Ag1.2.3 rat myeloma cell line. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
