Human CD49f molecules are integrin a6 chains that are expressed as heterodimers with CD29 (b1 integrin) or CD104 (b4 integrin) and function as adhesion receptors. CD49f is expressed on platelets, monocytes, T lymphocytes and thymoctyes. The ligands for CD49f/CD29 include laminin and invasion. CD49f/CD29 has been shown to associate with CD63. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:150 (10ug) Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 5 x 10e5 cultured Raji human tumor cells were washed and incubated 45 minutes on ice with 80ul of C2405-04R at a 1:50 dilution factor (10ug). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.32 log10 fluorescent units when compared to a mouse IgG1/PE negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20ul of 0.5mg/ml C2405-04N. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
