The CD52 antigen is a remarkably small peptide that is heavily glycosylated, and attached to the cell surface membrane via a GPI link. The apparent molecular mass of the antigen on SDS-PAGE is 25-29kD. CD52 is expressed at high density by lymphocytes, monocytes, eosinophils, thymocytes and macrophages. It is expressed by most lymphoid derived malignancies, although expression on myeloma cells is variable. Humanized versions of CAMPATH-1 specific antibodies are currently in clinical trials for the treatment of a range of lymphoid malignancies. Applications: Suitable for use in Cytotoxicity Assays, ELISA, Flow Cytometry, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (frozen): 1:20 Immunohistochemistry (paraffin): 1:20-1:40 Flow Cytometry: 1:20, use 10ul to label 1x10e6 cells in 100ul. Cytotoxic Assays: 50ug/ml (Use human serum as complement source) Optimal dilutions to be determined by the researcher. Recommended Positive Control Tissue: Tonsil Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
