Human CD53 is a tetraspan cell surface glycoprotein found on all hemopoetic cells except platelets and red blood cells. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 10ug/ml, 80ul labels 5x10e5 cultured human Jurkat cells. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 cultured human Jurkat cells per tube were incubated 45 minutes on ice with 80ul of C2408-21B at 10ug/ml. 2. Cells were then washed twice and incubated 45 minutes with Streptavidin(PE) (S7973-88P), 3. The cells were then washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 2.68 log10 fluorescent units when compared to a Mouse IgG2b/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20ul of 0.5mg/ml anti-CD53 antibody (C2408-21A). Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
