Human CD60 antigen is a glycolipid determinant found on about 30% of peripheral blood T cells. The glycolipid determinant is linked to a glycoprotein which can transduce activation signals. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50, 80ul labels 5x10e5 ficoll prepared human PBL cells. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 ficoll prepared human PBL were incubated 45 minutes on ice with 80ul of C2413-79B at a 1:50 dilution (10ug/ml). 2. Cells were then washed three times, fixed and analyzed by FACS. A 39% sub population of the cells stained positive with a mean shift of 2.3 log10 fluorescent units when compared to a Mouse IgM/FITC negative control at a similar concentration. 3. Binding was blocked when cells were pre incubated for 10 minutes with 20ul of 0.5mg/ml anti-CD60b antibody (C2413-79). Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.
