Collagen is an inert, rigid protein found predominantly in skin, ligaments, bones and teeth. Its most distinctive attribute, essential to a transmitter of mechanical force, is inelasticity. Its fundamental structural unit is tropo-collagen, a molecular rod about 2600A in length and 15A in diameter and 300,000 molecular weight. In tendons these macromolecules, grouped as collagen fibrils, run parallel to the axis. In skin the fibrils are interlaced and branched. Highly purified type II collagen. Used for the induction of collagen-induced arthritis (CIA) in experimental animals. Suitable for use as an antigen in immunological protocols. Solubility: Soluble in acidic buffer (max 4mg/ml). To dissolve in neutral buffer use 0.01M or 0.05M acetic acid to dissolve 1-4mg/ml collagen, then add neutral buffer (such as 2-10X Tris-NaCl, final concentration of sodium chloride: 0.15-0.2M), or dialyze against 1X neutral buffer at 4C. Note: If using a neutral buffer, Type II Collagen may not form stable gel with incubation at 37C. Additional components such as type I Collagen may be required. Note: To avoid fibril formation under neutral conditions, keep solution on ice. Storage and Stability: 4C or -20C at the dark. This product is stable at 4C for 2 years, and at -20C for 5 years as supplied. Collagen dissolved in acidic buffer is also stable at 4C for 1 month and -20C for 1 year, but will gradually degrade under neutral conditions.
