Collagen is an inert, rigid protein found predominantly in skin, ligaments, bones and teeth. Its most distinctive attribute, essential to a transmitter of mechanical force, is inelasticity. Its fundamental structural unit is tropo-collagen, a molecular rod about 2600A in length and 15A in diameter and 300,000 molecular weight. In tendons these macromolecules, grouped as collagen fibrils, run parallel to the axis. In skin the fibrils are interlaced and branched. Highly purified type II collagen may be used for assaying antibodies in sera from collagen-induced arthritis (CIA) animal models and humans by ELISA. Polymeric or fibrilar collagen can bind, nonspecifically, to immunoglobulins in serum samples, secondary antibodies and avidin-peroxidase creating high, false-positive reactions. To prevent this, and to obtain consistent assay results, USBio provides highly purified, polymeric-free, ELISA Grade Collagen. Applications: Suitable for use in ELISA. Other applications not tested. Recommened Dilution: It is recommended to dilute this product to 5-10ug/ml on ice with chilled Collagen-Dilution Buffer to coat ELISA plates just before use. For coating, use aliquots (do not keep diluted solutions). The optimum concentration of collagen to coat ELISA plates varies upon the type of plate used. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Light sensitive. Notes: 1. C7510-26 will gradually form polymeric collagen and degrade under neutral conditions. 2. Do not store diluted solutions of C7510-26.
