Collagenases degrade native helical collagen fibrils. The substrate, collagen, is the major fibrous component of animal extracellular connective tissue: skin, tendon, blood vessels, bone, etc. Suitable for use in cell dissociation procedures. Collagenase is prepared to contain lower proteolytic activity levels to limit damage to membrane proteins and receptors but with normal to above normal collagenase activity. Suitable for use in mammary primary cell isolation. Collagenase nomenclature: Type 1: Containing average amounts of assayed activities (collagenase, caseinase, clostripain and tryptic activities). It is generally recommended for fat, adrenal, and liver cells. Type 2: Containing greater clostripain activity. It is generally used for heart, bone, muscle, thyroid, cartilage, and liver cells. Type 3: Containing low proteolytic activity. It is usually used for mammary and fetal cells. Type 4: Containing low tryptic activity. It is commonly used for islets and other applications where receptor integrity is crucial. Activity: 100 units/mg dry weight. Units/mg Caseinase (dw): 50 Units/mg Clostripain (dw): 3 Units/mg Tryptic (dw): 0.3 Absorbance mg/ml 280 nm: As reported Unit Definition: 1 unit releases 1 µmole of L-leucine equivalents from collagen in 5 hours at 37C, pH 7.5. Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
