Collagenases are endopeptidases that digest native collagen in the triple helix region. Collagens are the major fibrous component of animal extracellular connective tissue. Bacterial collagenases differ from vertebrate collagenases in that they exhibit broader substrate specificity Unlike animal collagenases that split collagen in its native triple-helical conformation, bacterial collagenase is unique because it can degrade both water-insoluble native collagens and water-soluble denatured ones. It can attack almost all collagen types, and is able to make multiple cleavages within triple helical regions. Unit Definition: 1 unit releases 1umole of L-leucine equivalents from collagen in 5 hours at 37C, pH 7.5. Activity: 160units/mg dry weight Units/mg Caseinase (dw): As Reported Units/mg Clostripain (dw): As Reported Units/mg Tryptic (dw): As Reported Absorbance mg/ml 280nm: As Reported Activators: Zinc, Calcium Endotoxin EU/mg (dw): As Reported Applications: Suitable for use in pancreatic islet primary isolation and other applications where receptor integrity is crucial. Collagenase is prepared to contain lower tryptic activity levels to limit damage to membrane proteins and receptors but with normal to above normal collagenase activity. Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at 4C. Do not freeze. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
