The L858R mutation has been associated with non-small cell lung cancer patients who demonstrate clinical responsiveness to the tyrosine kinase inhibitor gefitinib (Iressa, ZD1839). Source: Recombinant human EGFR T790M L858R, aa668-1210, with N-terminal GST tag, expressed in baculovirus expression system. Molecular Weight: ~88kD Specific Activity: As reported Reaction Buffer: 50mM HEPES (pH7.5), 10mM MgCl2, 1mM EDTA, 0.01% BRIJ- 35 and 200uM of ATP. Substrate: 4uM Tyr peptide 4 from Invitrogen. Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Recommended Dilution: The enzyme reaction was carried out for 1h at room temperature in reaction buffer. Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
