IgA has a higher turnover rate (66mg/kg body weight/day) than all other immunoglobulins combined. In secretions the majority of IgA (S-IgA) exists as a dimer (2x150kD units) linked together by J-chain (MW. 16kD) and protected from a proteolytic digestion by secretory component (70kD). Unlike serum, where IgA1is the dominant subclass, in secretions the proportion of IgA1 and IgA2 varies (IgA2 has increased resistance to proteolytic degradation). Free secretory component is also found in varying concentrations in body fluids, as is secretory IgM, which is similar to the serum form, but has secretory component attached. S-IgA forms a first line of protection on mucosal surfaces where it inhibits uptake of antigen and interferes with microorganism adherence to the mucosal epithelium. Cells such as macrophages, neutrophils, hepatocytes and natural killers have surface receptors for IgA. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: ELISA: 1:56,000 Western Blot: 1:1000-1:5000 Immunohistochemistry: 1:500-1:2500 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. Form: Supplied as a lyophilized powder from PBS, pH 7.2, 10mg/ml BSA (IgG and protease free), 0.01% gentamicin sulfate. Labeled with Horseradish Peroxidase (HRP). Reconstitute with 1ml sterile ddH2O.
