Interleukin-10 (IL-10), originally known as Cytokine Synthesis Inhibitory Factor (CSIF), shares over 80% sequence homology with the Epstein-Barr Virus protein BCRFI. The reported biological activities of IL-10, which may be interrelated, include inhibition of macrophage-mediated cytokine synthesis, suppression of the delayed-type hypersensitivity response, and stimulation of the Th2 cell response which results in elevated antibody production. Human IL-10 is active on murine cells. Murine IL-10 is inactive on human cells. Recombinant human IL-10 is an 18.6kD protein with 161 amino acid residues. The ED50 was found to be 2ng/ml, corresponding to a specific activity of 5x10e5 units/mg. Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20C. Reconstitute with sterile 5mM Tris, pH 7.2. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Purity:
98% (SDS-PAGE and HPLC). Endotoxin: 0.1ng/ug
Form:
Supplied as a lyophilized powder from 5mM sodium phosphate, pH 7.2, without additives. Reconstitute with 5mM Tris, pH 7.2 to a concentration of 0.1-1mg/ml.
