Involucrin from human keratinocytes. MW of Antigen: 66-170kD. Cellular Localization: Cytoplasmic. The epitope maps between residues 421-568 of human involucrin.
Involucrin is expressed in a range of stratified squamous epithelia, including the cornea. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures it is expressed by all cells that have left the basal layer. Involucrin expression is abnormal in squamous cell carcinomas and premalignant lesions and is reduced in severe dysplasias of the larynx and cervix. Applications: Suitable for use in ELISA, Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunoprecipitation: 2ug/mg protein lysate (Denatured verified, Use Protein G). Western Blot: 1-2ug/ml for 2 hours at RT. Immunohistochemistry (Formalin/paraffin): 1:100-1:200 for 20 minutes at RT. For staining formalin-fixed tissues, digest sections with trypsin at 1mg/ml PBS, for 10 minutes at 37C or Protease XXV at 1mg/ml PBS, for 5 minutes at 37C. Note: Enzyme digestion is better than citrate buffer based epitope unmasking. Optimal dilutions to be determined by the researcher. Positive Control: LS174T cells and skin Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
