Full length sequence of mouse p42 mitogen activated protein kinase (MAPK2) fused to glutathione S-transferase (GST) at the amino terminus and expressed in E. coli. The GST moiety can be removed by thrombin treatment if needed. An important convergence point involved in the signal transduction pathways of many different growth factors, hormones, and cytokines is a family of 41-44kD serine/threonine kinases collectively called either MAPKs (for mitogen-activated protein kinases) or Erks (for extracellular-regulated kinases). MAPK is activated by sequential phosphorylation on both tyrosine and threonine residues by either the dual tyrosine/threonine kinase MEK (MAPK Kinase) alone or by MEK in conjunction with a yet undescribed kinase. The serine/threonine kinases Raf and MEKK (MAPK Kinase Kinase) phosphorylate and activate MEK, in turn, during intracellular signaling. Activation of MAPK is directly regulated by a specific MAPK phosphatase and indirectly regulated by protein kinase A activation that results in inhibition of Raf activity in mammalian cells. Specific Activity: ~1000units/mg, where 1 unit = 1nmole of phosphate incorporated into 0.33mg/ml myelin basic protein/min at 30C with a final ATP concentration of 100uM. Molecular Weight: ~67.8kD Quality Control Testing: Protein Kinase Assay: 10-100ng of this lot successfully phosphorylated MBP in the radioactive kinase assay described in the protocol. MBP was captured using Streptavidin- Agarose. A previous lot successfully phosphorylated 32P-labeled biotinylated MBP. MS Tryptic Fingerprint: Confirmed identity as MAPK2 with 41% sequence coverage of the translated swquence listed in protocol. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
