| The protein encoded by this gene is a substrate for protein kinase C. It is localized to the plasma membrane and is an actin filament crosslinking protein. Phosphorylation by protein kinase C or binding to calcium-calmodulin inhibits its association with actin and with the plasma membrane, leading to its presence in the cytoplasm. The protein is thought to be involved in cell motility, phagocytosis, membrane trafficking and mitogenesis. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilutions: Western Blot: 1:500-1:1000. Detects MARCKS in a bovine brain extract. Bovine brain extract was resolved by electrophoresis, transferred to nitrocellulose and probed with M2375 (1:500). Proteins were visualized using an IgG (HRP) goat anti-mouse* and a chemiluminescence detection system. Immunoprecipitation: Precipitates MARCKS proteins from bovine brain extracts. Used mainly for the evaluation of MARCKS protein phosphorylation in intact human cells. Determined by immunoprecipitation of 32P-labeled protein as an indicator of protein kinase C activation in cells. Optimal dilutions to be determined by the researcher. *Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
