The scavenger receptors (SRs) expressed by macrophages are thought to play an important role in the immune response against bacteria by mediating ligand binding and phagocytosis. An important family of receptors involved in these processes are the class A macrophage scavenger receptors (SR-A). There are three classes of SRs, namely A, B and C. SR-A, type I and type II, are homotrimeric glycoproteins composed of three 77kD monomers. MARCO is a member of this class A of scavenger receptors. The molecular structure of MARCO resembles that of SR-AI, containing a triple-helical collagenous domain and a scavenger receptor cysteine-rich (SRCR) domain at the C terminus. MARCO is expressed by distinct populations of macrophages in the spleen and lymph nodes. It is rapidly induced on macrophages in other tissues (e.g. Liver Kupfer cells) during infection or LPS treatment. MARCO seems to play an important role in the lungs response to inhaled particles and airborne pathogens. Applications: Suitable for use in Immunofluorescence, Immunoprecipitation, Flow Cytometry, Immunohistochemistry and Western Blot (Non-reducing). Also suitable for Functional Studies where the antibody blocks human alveolar macrophages binding to unopsonized particles. Other applications not tested. Recommended Dilutions: Immunofluorescence: 0.6ug/ml stained alveolar macrophages and transfected CHO cells Immunoprecipitation: Immunoprecipitates MARCO as 60 and 50kD protein from lysates obtained from COS cells transfected with human MARCO. Flow Cytometry: 1:50. Weakly stains alveolar macrophages by recognizing the extracellular domain of MARCO. Immunohistochemistry (Frozen): 5ug/ml. Tissue sections can be fixed in acetone or 2% paraformaldehyde. Optimal dilutions to be determined by the researcher. Positive Control: Human alveolar macrophages Storage and Stability: May be stored at 4C. For long-term storage, aliquot and store at 4C. Do not freeze. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
