MELAN-A/MART-1 is a melanocyte differentiation marker. MELAN-A is expressed by a large proportion of melanoma tumors, both melanotic and amelanotic and by melanoma cell lines. It is present in melanocytes of normal skin and retina, nevi and in more than 85% of melanomas. MELAN-A/MART-1 is not expressed by non-melanoma malignancies. It is recognized by autologous cytotoxic T lymphocytes. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. Seven other melanoma-associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-1, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Applications: Suitable for use in Immunoprecipitation, Western Blotting, and Immunohistochemistry (frozen & formalin/paraffin). Other applications have not been tested. Recommended Dilution: Immunoprecipitation: Use Protein G, 2ug antibody/mg protein lysate Western Blotting: 1ug/ml for 2 hours at RT. Immunohistochemistry: 1-2ug/ml for 30 min. at RT Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min., followed by cooling at RT for 20 min. Optimal dilutions to be determined by the researcher. Positive Control: CaC1 melanoma cells and melanomas. Cellular Localization: Cytoplasmic. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
