Purified human skin chymase. Cellular Localization: Cytoplasmic
Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of chymase is estimated to be found in mast cells of skin and lung and suggests this enzyme plays a major role in mast cell mediated events. Human mast cell chymase is considered to be an important marker of mast cells as well as an important mediator of inflammation. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry (Formalin/paraffin): 1:2000 for 20 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at RT for 20 minutes. Optimal dilutions to be determined by the researcher. Positive Control: Tonsil Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
