Mast Cell Protease 11 (MCP-11) encoded by the Prss34 gene is a secreted trypsin-like serine protease. It consists of a signal peptide (aa 1 to 19), a pro region (aa 20 to 34), and a catalytic domain (aa 35 to 318). The amino acid sequence of mouse MCP-11 is 84% and 60% identical to that of rat and dog. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Direct ELISA: 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse MCP-11. The detection limit for rmMCP-11 is approximately 10 ng/well. Western Blot: 1-2ug/ml with the appropriate secondary reagents to detect mouse MCP-11. Using a colorimetric detection system, the detection limit for rmMCP-11 is approximately 25 ng/lane under non-reducing and reducing conditions. Chemiluminescent detection with WesternGlo(TM) Chemiluminescent Detection Substrate will increase sensitivity by 5 to 50 fold. Immunoprecipitation: 25ug/ml to immunoprecipitate rmMCP-11 from conditioned cell culture medium. The recovered rmMCP-11 can be detected in Western blots using a polyclonal antibody. Optimal dilutions to be determined by the researcher. Storage and Stability: For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
