Tryptase is the most abundant secretory granule-derived serine proteinase contained in mast cells. Tryptase has recently been used as a marker for mast cell activation and is involved with allergic response. Tryptase may act as a mitogen for fibroblast lines. Elevated levels of serum tryptase occur in both anaphylactic and anaphylactoid reactions, but a negative test does not exclude anaphylaxis. Applications: Suitable for use in Western Blot, Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: For optimal results, tissues or cytospin preparations should be fixed in Carnoys Fluid (60% ethanol, 30% chloroform, 10% glacial acetic acid). Not recommended for use on formaldehyde fixed tissue. May be used on thin sections prepared for ultrastructural analysis after fixation of tissue in a modified Karnovskys reagent (Lab Invest. (1988). 58:682-691). Western Blot: detected tryptase in 10ug of human fetal skin lysate. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
