Metabotropic glutamate receptors have been classified into three groups. In general, class I receptors (mGluR1 and mGluR5) mediate the excitation of neurons, whereas activation of class II (mGluR2 and mGluR3) and class III (mGluR4a, mGluR7a, mGluR7b and mGluR8) results in a depression of synaptic transmission. Class I mGluRs are localized in the postsynaptic membrane and are positively coupled to phospholipase C (PLC). Class II and class III mGluRs localized to the presynaptic membrane and are negatively coupled to adenylate cyclase. It has been speculated that mGluR2 and/or mGluR3 may play a role in the modulation of the mossy terminal function.
