MGMT (O6-methylguanine-DNA methyltransferase). Many tumor promoting/causing agents (BCNU, CCNU, DTIC, procarbazine, temozolomide, etc) generate lethal cross-links at the O6-alkylguanine position in DNA. MGMT removes alkyl adducts from the O6-position of guanine in DNA (prior to cross-link formation) to its cysteine residue, thereby irreversibly inactivating MGMT. Inactivated MGMT is rapidly degraded. Therefore, MGMT levels serve as an indicator of DNA damage. Tumors with high level of MGMT are likely to be drug resistant and low or no MGMT may make the tumor more responsive to chemotherapy. In normal cells, abnormally low level of MGMT may make normal cells/tissue more susceptible to tumor promoting/alkylating agents. Biological Activity: Biologically inactive Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: Heat once and load 10ul/lane and visualize with M3884-91C & M3884-91E. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
