Agent Gram-negative pleomorphic cells, spherical to ovoid shaped, single tripled-layered membrane but lacking true cell wall. Causes lower respriratory infections, less often a pharyngitis progressing to bronchitis or pneumoniae, gradual onset with headache, malaise, paroxysmal cough, substernal pain, leukocytosis in some cases. Duration varies from a few days to a month or more. Low fatality. Inactivation: Ether Inactivation Applications: Suitable for use in IgG and IgM detection in assays which include EIA with polystyrene and latex solid phases. This antigen preparation should be sonicated immediately prior to use to ensure that the preparation is uniform. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Preparation: Mycoplasma organisms are harvested by centrifugation, washed in PBS and extracted using ether. Residual ether is removed from the aqueous phase by bubbling with nitrogen gas. Resulting antigen preparation contains soluble Mycoplasma components in PBS. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
