Control Peptide for N2017-23 (antiserum) and N2017-30 (affinity purified antibody). Na+/H+ exchangers (NHE) of mammalian cells are plasma membrane intrinsic proteins mediating exchange of N+ and H+ ions in various tissues. The NHE catalyzes the electroneural transport of extracellular Na+ for intracellular H+. They play a major role in regulation of intracellular pH (pHi) in addition to transcellular absorption of Na+, cell volume regulation and possibly in cell proliferation. These primary functions of the Na+/H+ exchanger have been related to many pathophysiological states, including hypertension, organ growth and hypertrophy, regression of cancer and renal intestinal disorders. At least 7 NHE isoforms (NHE1-7) have been cloned so far. They are all similar in their primary structure and predicted to have 10-12 transmembrane domains. The C-terminal domain of NHEs are predicted to be intracellular. NHE3 (rat, 831aa, human chromosome 5p15.3) is involved in trans-epithelial sodium absorption. The NHE3 mRNA is found in kidney cortex, medulla, jejunum, ileum, colon and stomach. Applications: Suitable for use in ELISA and Antibody Blocking. Not suitable for use in Western Blot. Other applications not tested. Recommended Dilution: ELISA: 50-100ng control peptide/well. Antibody Blocking: 5-10ug per 1ul N2017-23 (antiserum) or per 1ug N2017-30 (affinity purified antibody). Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
