ADPSugar Pyrophosphatase (NUDT5)is a member of the nudi x super family of enzymes. Members of this family are pyrophosphohydrol gases that act upon substrates with the general structure of a nucleoside(Nu) diphosphate (di) linked to another moiety, X (NDPX) to yield NMP plus PX (1).Human NUDT5 is a homodimeric enzyme present in the cytosol of most cell types (2). Glu166 and three magnesium ions are important for stabilizing the transition state during the hydrolysis of ADPR (3) . NUDT5 has been suggested to play a role in regulating the intracellular levels of ADPR by NO activation through ADP ribosyl at ion at cysteine residues of the enzyme in macrophages (4). It also may play defensive role against the mutagenesis induced by oxidized deoxyribonucl eosides (5,6). Source: Recombinant corresponding to Glu2-Phe219 from human ADP-Sugar Pyrophosphatase/NUDT5, N-terminal, Met, 6-His tag, expressed in E. coli. Molecular Weight: ~25kD Biological Activity: Measured by its ability to hydrolyze ADP-ribose to AMP and ribose-5-phosphate. The specific activty is >1,000 pmoles/min/ug, as measured under the described conditions. Endotoxin Level: <1EU/ug (LAL method) Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing.. Store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
