Synthetic peptide corresponding to amino acids at C-terminus of human PHAP I (1). Apoptosis is related to many diseases and development. Caspase-9 plays a central role in cell death induced by a variety of apoptosis activators. Cytochrome c, after being released from mitochondria, binds to Apaf-1, which forms an apoptosome that in turn binds to and activates procaspase-9. Activated caspase-9 cleaves and activates the effector caspases (caspase-3,-6 and -7), which are responsible for the proteolytic cleavage of many key proteins in apoptosis. The tumor suppressor putative HLA-DR-associated proteins (PHAPs ) were recently identified as important regulators of mitochondrion apoptosis (1). PHAP appears to facilitate apoptosome-mediated caspase-9 activation and to stimulates the mitochondrial apoptotic pathway. PHAP was also shown to oppose both Ras and Myc-mediated cell transformation. Applications: Suitable for use in ELISA and Antibody Blocking . Other applications not tested. Recommended Dilution: Preadsorption Control: 1ug peptide per 1ug aP3368-02 for 30 min at 37°C. Optimal dilutions to be determined by the researcher. Storage and Stability: For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
