Synthetic peptide corresponding to amino acids close to C-terminus of human PHAP I (1). This sequence is identical between human and rat PHAP I. Corresponding antibody detects PHAP I. Species Crossreactivity: human and rat PHAP I. Apoptosis is related to many diseases and development. Caspase-9 plays a central role in cell death induced by a variety of apoptosis activators. Cytochrome c, after released from mitochondria, binds to Apaf-1, which forms an apoptosome that in turn binds to and activate procaspase-9. Activated caspase-9 cleaves and activates the effector caspases (caspase-3,-6 and -7), which are responsible for the proteolytic cleavage of many key proteins in apoptosis. The tumor suppressor putative HLA-DR-associated proteins ( PHAPs ) were recently identified as important regulators of mitochondrion apoptosis (1). PHAP appears to facilitate apoptosome-medicated caspase-9 activation and to stimulate the mitochondrial apoptotic pathway. PHAP was also shown to oppose both Ras and Myc-mediated cell transformation. Applications: Suitable for use in ELISA, Western Blotting. Other applications not tested. Recommended Dilution: Western Blotting: The peptide is used for blocking the activity of PHAP I antibody. It usually blocks the antibody activity completely in Western blot by incubating the peptide with equal volume of antibody for 30 min at 37°C. Optimal dilutions to be determined by the researcher. Corresponding Antibody: P3368-03 Positive Control: Raji cell lysate Storage and Stability: Lyophilized powder may be stored at 4C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
