Synthetic peptide corresponding to amino acids at C-terminus of human PHAP I (1). Corresponding antibody detects PHAP I and PHAP III Species Crossreactivity: human, mouse and rat. Apoptosis is related to many diseases and development. Caspase-9 plays a central role in cell death induced by a variety of apoptosis activators. Cytochrome c, after released from mitochondria, binds to Apaf-1, which forms an apoptosome that in turn binds to and activate procaspase-9. Activated caspase-9 cleaves and activates the effector caspases (caspase-3,-6 and -7), which are responsible for the proteolytic cleavage of many key proteins in apoptosis. The tumor suppressor putative HLA-DR-associated proteins ( PHAPs ) were recently identified as important regulators of mitochondrion apoptosis (1). PHAP appears to facilitate apoptosome-medicated caspase-9 activation and to stimulate the mitochondrial apoptotic pathway. PHAP was also shown to oppose both Ras and Myc-mediated cell transformation. Applications: Suitable for use in ELISA, Western Blotting. Other applications not tested. Recommended Dilution: Western Blotting: The peptide is used for blocking the activity of PHAP antibody. It usually blocks the antibody activity completely in Western blot by incubating the peptide with equal volume of antibody for 30 min at 37°C. Optimal dilutions to be determined by the researcher. Corresponding Antibody : P3368 Positive Control: Raji cell lysate Storage and Stability: May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20C or colder. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
