This plasmin substrate is chemically and functionally identical to Chromogenix S-2251. Each 25mg of substrate is lyophilized with 60mg mannitol as a bulking agent. Formula: H-D-Val-Leu-Lys-pNA2HCl Activity: The method for the determination of activity is based on the difference in absorbance between the pNA formed and the original substrate. The rate of pNA formation, i.e. the increase in absorbance per second at 405nm, is proportional to the enzymatic activity and is determined with a spectrophotometer. Stock Solution: 10mmol/L in 1mM HCl Working Solution: 200umol/L in 0.1M HEPES, 0.1M sodium chloride, pH 7.4 Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 6 months after receipt. Reconstitute with sterile buffer. Aliquot to avoid repeated freezing and thawing. Store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
