One unit is defined as the amount of enzyme that will phosphorylate 1 nmol of casein substrate per minute at pH 7.4 and 30C. Assay Buffer: 10mM HEPES, pH 7.4, 5mM MgCl2, 10uM MnCl2, 1mM DTT, 3uM Na-orthovanadate, 0.4mM cold ATP/ 2uCi [g-32P]ATP and 0.5ug/ul alpha-casein substrate.
