Preparation: Pure Streptavidin is reacted with Succinimidyl 4- (N-maleimido-methyl) cyclohexane-1- carboxylate (SMCC). R. Phycoerythrin is reacted with N-succinimidyl 3-(2-pyridyldithio) propionate (SPDP) and activated by reduction with dithiothreitol. The two substitiuted proteins are reacted together to give covalent conjugates, which are selected by medium pressure liquid chromatography on gel filtration columns of AcA34 ultrogel. Conjugates show negligible non-specific binding to non-biotinylated macromolecules, and therefore gives very low backgrounds. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: Neat. Use 10ul to label 10e6 cells in 100ul. Optimal working dilutions to be determined by researcher. Storage and Stability: Store product at 4C before and after reconstitution. DO NOT FREEZE! Lyophilized and reconstituted products are stable for 12 months at 4C after receipt. Reconstitute lyophilized powder with sterile 1ml dH2O prior to use and store remainder as an undiluted liquid. Caution: PE conjugates are sensitive to light. For maximum recovery of product centrifuge the original vial prior to removing the cap.
